چکیده

Bovine milk samples were collected from cases of clinical and sub-clinical mastitis, respectively and examined bacteriologically and by simplex and multiplex PCR assays for detection of Staphylococcus aureus, Escherichia coli and Streptococcus agalactiae. Escherichia coli was the most common bacteria detected in the collected samples from clinical mastitis cases followed by Staphylococcus aureus and Streptococcus agalactiae; while Staphylococcus aureus was the most common bacteria followed by Escherichia coli and Streptococcus agalactiae in samples collected from sub-clinical mastitis cases. Compared with the cultural method, the simplex and multiplex PCR assays are less time consuming. It took less than 24 hours to be completed, while identification of bacteria to the species levels by conventional microbiological and biochemical methods required more than 72 hours. In conclusion, simplex and multiplex PCR assays can be used as a rapid, sensitive and specific routinely diagnostic tool to detect the presence of Staphylococcus aureus, Escherichia coli and Streptococcus agalactiae in milk samples.

Field comparison of real-time polymerase chain reaction and bacterial culture for identification of bovine mastitis bacteria.

Fast and reliable identification of the microorganisms causing mastitis is important for management of the disease and for targeting antimicrobial treatment. Methods based on PCR are being used increasingly in mastitis diagnostics. Comprehensive field comparisons of PCR and traditional milk bacteriology have not been available. The results of a PCR kit capable of detecting 11 important etiologi...

A polymerase chain reaction based study on the subclinical mastitis caused by Streptococcus agalactiae, S. dysgalactiae and S. uberis in cattle in Ahvaz

Streptococcus agalactiae , S. dysgalactiae and S. uberis are Gram-positive bacterial pathogens that affect cows in dairy herds. These are considered as the major causes of economic losses of dairy producers without a control program. To reveal the role of these agents in inducing subclinical mastitis in dairy cattle in Ahvaz, 100 CMT-positive milk samples and 20 CMT-negative milk samples were c...

Direct Identification of Major Pathogens of the Bubaline Subclinical Mastitis in Egypt using PCR

The present study aimed to evaluate the PCR assays (using Universal and specific primers) for detection of the major pathogens of bubaline subclinical mastitis directly from 160 buffaloes milk samples compared to bacterial examination. The bacteriological examinations showed incidence of 23.75%, 15.62%, 3.12% and 0.62% for E. coli, S. aureus, S. agalactiae and S. dysgalactiae respectively. PCR ...

Detecting Virulence Factors and Antibiotic Resistance Pattern of Trueperella Pyogenes Isolated from Bovine Mastitic Milk

Backgrounds and Aims: Mastitis is a mammalian disease which is considered important due to its potential economic damages. Trueperella pyogenesis is one of the important opportunistic pathogens of the mammary glands of cattle. This bacterium can produce acute mastitis infection in dairy cattle. In fact, this bacterium has several virulence genes which contribute to its pathogenicity. Therefore,...
